چکیده

IKK/NF-kB signalling controls inflammation and cell death is important to maintain tissue homeostasis. Keratinocyte-specific deletion of IKK2 (IKK2E-KO) leads TNFR1-dependent skin in mice. IKK2E-KO mice showed increased numbers apoptotic necrotic keratinocytes, upregulation inflammatory cytokines mediators the epidermis. We proposed that keratinocyte could trigger inflammation. Receptor Interacting Protein Kinase (RIPK1) induces via kinase activity-dependent functions. To address kinase-dependent role RIPK1 mice, we generated expressing inactive (IKK2E-KO; Ripk1D138N/D138N). observed IKK2E-KO;Ripk1D138N/D138N did not develop lesions, demonstrating an essential activity further dissected whether apoptosis or necroptosis drives by crossing them lacking RIPK3 MLKL, which are necroptosis. found IKK2E-KO;Ripk3-/- IKK2E-KO;Mlkl-/- were strongly protected from lesion development, showing triggering dissect different components NF-kB pathway regulating homeostasis, with combined deficiency subunits, cRel RelA a regulatory subunit IKK complex, NEMO (NEMOE-KO) Both, cRel/RelAE-KO NEMOE-KO developed inflammation, however, RIPKI but These results highlight differential control homeostasis through unravel potential unknown controlling

Computational Models of the NF-KB Signalling Pathway

In this review article, we discuss the current state of computational modelling of the nuclear factor-kappa B (NF-κB) signalling pathway. NF-κB is a transcription factor, which is ubiquitous within cells and controls a number of immune responses, including inflammation and apoptosis. The NF-κB signalling pathway is tightly regulated, commencing with activation at the cell membrane, signal trans...

Spiky oscillations in NF-kB signalling

The NF-κB signalling system is involved in a variety of cellular processes including immune response, inflammation, and apoptosis. Recent experiments have found oscillations in the nuclearcytoplasmic translocation of the NF-κB transcription factor [Hoffmann et al. (2002) Science 298, 1241; Nelson et al. (2004) Science 306, 704.] How the cell uses the oscillations to differentiate input conditio...

Alteration in Inflammation-related miR-146a Expression in NF-KB Signaling Pathway in Diabetic Rat Hippocampus.

PURPOSE The purpose of the present study is to evaluate the expression of miR-146a gene, its adaptor genes (TRAF6, NF-KB, and IRAK1), and possible changes in the cellular signaling pathway in diabetic hippocampus tissue. METHODS Male Sprague-Dawley rats are randomly selected and divided into control and diabetic (n=6) groups. Diabetes induced by the single-dose injection of nicotinamide [110 ...

The protective effect of nano-curcumin in experimental model of acute pancreatitis: The involvement of TLR4/NF-kB pathway

Objective(s): The objective of the present study is to explore whether Nanocurcumin improves pancreatic inflammation through the inhibition of the TLR4/NFkB signaling pathway in cerulein-induced acute pancreatitis. Methods: Acute pancreatitis was induced by five intraperitoneal (i.p.) injection of cerulein (50 μg/kg) with 1h intervals. Vehicle and nanocurcumin (100mg/kg/day) were given to the a...
